What belongs in a probiotic pouch acceptance specification?

Define the strain, viable-count basis, method, storage and expiry decision before agreeing a headline number.

Make the identity unambiguous

An acceptance specification begins with the exact material. Record the strain designation, supplier, composition, carriers and handling conditions. A species name may not connect a material to the evidence cited for a particular strain. The formulation team also needs to know which changes require supplier or brand approval.

Ask whether the project concerns a live culture or a non-live microbial preparation. A postbiotic direction cannot be labeled as live probiotic CFU. It has different identity and evidence requirements. C24 in our concept register exists to make that distinction explicit during the early discussion.

Define the count and its timing

A viable count needs a method, sample basis and time point. State whether the result is per gram, per pouch or per retail unit and how that basis is calculated. Define the recovery procedure from the final matrix. A supplier powder count and a finished-pouch count are not automatically equivalent.

Release and end-of-shelf-life requirements are separate. ISAPP guidance highlights strain identity and viable numbers through shelf life in probiotic labeling. For a pouch, the technical team must determine how to apply the relevant evidence and testing to the actual formulation, use and market. A label number without this context is difficult to interpret.

Connect the specification to the pack

The candidate can, closure and seal are part of the stability system. Record the intended storage statement and whether repeated opening needs evaluation. A culture that remains viable in a sealed ingredient sachet may behave differently after mixing, pouch filling and packaging.

The study plan should identify formula and pack versions, conditions, time points, analytical methods and acceptance criteria. Stability limits should follow the evidence and project assessment. They should not be selected solely to fit a desired commercial shelf life.

Agree what happens when a result fails

An acceptance specification also needs a decision process. Define who reviews an unexpected count, whether a method issue must be investigated, which retained samples are available and how the batch or prototype is controlled during review. Avoid automatically retesting until a passing value appears.

The contract should identify responsibility for data access, change approval, release and label updates. A clear specification is useful to both buyer and manufacturer because it separates a measurable requirement from an unresolved research question. Final acceptance criteria and market claims remain project-specific.

Continue the project review

Use the specification worksheet to retain the relevant version, owner and decision. For physical or sensory observations, the sample evaluation guide helps keep comparison conditions consistent. Send a concise project brief when you are ready to define the work.

Project questions

Does this guide establish a finished-product result?

No. It sets out a development and procurement method. Product-specific data, methods and responsible review are needed before a specification or claim is approved.

Which version should the records refer to?

Identify the formula, pouch, packaging and method versions together. Record the sample or batch code and date so observations and decisions can be traced to the material actually evaluated.
